چکیده

Colloidal silver products are sold for a wide range of disinfectant and health applications. This has increased the potential human exposure to nanoparticles (AgNPs) ions (Ag+), which oral ingestion is considered be major route exposure. Our objective was evaluate compare toxicity two commercially available colloidal on intestinal epithelial models under realistic conditions. Mesosilver™ AgC were characterized concentration between 0.1 12 ?g/mL chosen. Caco-2 cells vs. co-culture mucus-secreting HT29-MTX (90/10) used. Repeated carried out determine cell viability over 18 days differentiation in 24-well plates. Selected concentrations (0.1, 1, 3 ?g/mL) tested cultured E-plates Transwells with same repeated regimen, impedance, trans-epithelial electrical resistance (TEER), respectively. Silver uptake, intracellular localisation, translocation determined by CytoViva™, HIM-SIMS, ICP-MS. Genotoxicity acutely-exposed proliferating ?H2AX immunofluorescence staining. given AgC, composed solely ionic silver, generally exerted more toxic effects than Mesosilver™, contains mix AgNPs silver. Due its patchy structure, presence mucus Caco-2/HT29-MTX only slightly mitigated deleterious viability. Increased genotoxicity observed cells. similar. In conclusion, show different levels gut due forms distinct (AgNPs and/or Ag+) contained within. study highlights applicability high-resolution (chemical) imaging detect localize provides insights into uptake mechanisms, fate cellular effects.

Evaluation of Iron Bioavailability in Caco-2 cell Culture Model: Modification of the Original Method

Background and Objectives: In the original in vitro method for evaluation of iron bioavailability using caco-2 cell model, ferritin formation by cells is considered as a functional indicator of both absorption and utilization of iron. However, as iron is measured in the tested material and then ferritin concentration is measured in caco-2 cell lysate, it is almost impossible to have a concrete ...

Nephrotoxicity comparison of two commercially available generic vancomycin products.

To date, no comparative clinical studies have investigated the effects of different vancomycin products on nephrotoxicity. The objective of this single-center, retrospective, matched-cohort study was to investigate the impact of two different vancomycin products on the development of nephrotoxicity. The study population included adults receiving a single vancomycin product, from either Pfizer o...

In-vitro cellular uptake and transport study of 9-nitrocamptothecin PLGA nanoparticles across Caco-2 cell monolayer model

The uptake and transport of 9-nitrocamptothecin (9-NC), a potent anticancer agent, across Caco-2 cell monolayers was studied as a free and PLGA nanoparticle loaded drug. Different sizes (110 to 950 nm) of 9-nitrocamptothecin nanoparticles using poly (lactic-glycolic acid) were prepared by via the nanoprecipitation method. The transport of nanoparticles across the Caco-2 cell monolayer as a func...
